Survey research fields adjacent to LHON for techniques, technologies, or approaches that could be applied to treatment or cure.

Background

The most exciting LHON research often comes from unexpected places. The 2026 TALED (Transcription Activator-Like Effector-based DNA-binding protein) mitochondrial gene-editing breakthrough was adapted from a technology originally developed for other applications. Current promising approaches include:

  • Gene therapy - Lenadogene Nolparvovec (approved), NR082 by Neurophth (Phase 3)
  • Mitochondrial gene editing - TALED-based direct correction of mtDNA mutations
  • Allotopic gene expression - Re-coding faulty mitochondrial genes for nuclear delivery
  • Neuroprotection - Idebenone and next-generation neuroprotective agents
  • Stem cell therapy - Optic nerve regeneration (preclinical)
  • Mitochondrial transplantation - Replacing dysfunctional mitochondria (early research)

What Needs to Be Done

Look beyond current LHON research to find innovations that could be applied:

  1. Adjacent gene therapies - What breakthroughs in other retinal diseases could apply to LHON?
  2. Mitochondrial medicine advances - New approaches from the broader mitochondrial disease field
  3. Neuroscience innovations - Optic nerve regeneration, neuroprotection strategies
  4. AI-driven drug discovery - Computational approaches to finding new therapeutic targets
  5. Cross-disciplinary opportunities - Nanotechnology, optogenetics, synthetic biology

Why This Matters

The cure for LHON may already exist in a different field. By systematically surveying adjacent research, we can identify opportunities that specialists focused narrowly on LHON might overlook.

Submit your results by opening a GitHub issue with the label discover-solutions. Include your analysis with feasibility assessments and source papers.

Success Criteria

  • Identify at least 5 promising approaches from outside traditional LHON research
  • For each approach: technology name, current application, potential LHON application, key papers, feasibility assessment
  • Include analysis of the 2026 TALED mitochondrial gene-editing breakthrough and its implications
  • Assess each approach on a scale of feasibility, timeline, and potential impact
  • Identify researchers in adjacent fields who could be recruited to LHON research
